What could have caused Penicillin Hypersensitivity?

A patient who had already taken amoxicillin several times but had no adverse drug reaction. Recently he developed a rash after taking amoxicilin. Why did this happen? He didn't take any other drugs with amoxicillin. Is this penicillin hypersensitivity? If so, why didn't it occur early when he took amoxicilin1st or 2nd time?

    If your friend has taken amoxicillin many times before and this was the first time an allergic reaction has occurred I would be specious if the reaction was from the amoxicillin. Was he taken anything else? What did the rash look like? There are a whole bunch of questions that need to be answered.
    In any case he should use caution before he attempts this drugs again.
    Better yet he should either get skin tested for penicillin allergy or not use this class of antibiotics again.

    I agree with Dr Hayes, in that, if your friend has taken amoxicillin "many" times before, then the reaction is unlikely to be due to amoxicillin. I also agree with allergy sensitivity testing. Let me address the last part of your question. Allergies occur, when our body recognizes a foreign protein and develops an immune response. So you cant have a true allergic reaction with the first dose. However, you can develop it over a 7-10 day course and develop the reaction to the later doses.

    Dr.Hayes and Dr.Lane,the case was diagnosed as a case of hypersensitivity reaction caused by amoxicillin and he is now taking another group of drug.Actually the patient is a sufferer of chronic tonsillitis and was given amoxicillin this time as preparation for tonsillectomy.Before that he took amoxicillin several times for episodic attack of tonsillitis.But my point is that....is it possible to develop hypersensitivity reaction to any drug previously nonhypersensitive though he took it several times ...by any sorts of mutation or something else?
